Abstract

Pentadecapeptide BPC 157 modulates sythesis of NO, has antiarrhythmic effect and protective effect on endothelium. This study was designed to investigate its effects in monocrotaline induced pulmonary hypertension in rats. It included 13 groups by 6 rats (body weight 150-200 g). Pentadecapeptide BPC 157 was given daily, either in drinking water or intraperitoneally, in two different dosages (10 μg/kg i 10 ng/kg), in cotreatment protocol day 1 - 29, while in posttreatment protocol therapeutic intervention with pentadecapeptide BPC 157 was started when the development of pulmonary hypertension had already commenced and was upheld day 15 - 29. In control groups a marked right heart hypertrophy was evident, and massive thickening of the precapillary artery smooth muscle layer was histologically apparent, with cilincal deterioration due to pulmonary hypertension during the 4. week after MCT injection, and some animals dying during this period because of right heart failure. In cotreatment protocol pentadecapeptide BPC 157 consistently prevented in all application modalities the development of pulmonary hypertension with all its manifestations, while in posttreatment therapeutic protocol BPC 157 reversed also in all application modalities the already established PAH. We conclude that pentadecapeptide BPC 157 prevents and attenuates MCT-induced pulmonary hypertension and cor pulmonale in rats.

Abstract in Croatian

Pentadekapeptid BPC 157 modulira sintezu NO, ima antiaritmijski učinak i protektivno djelovanje na endotel. U ovoj studiji cilj je bio ispitati njegov učinak u štakorskom modelu plućnog srca induciranog monokrotalinom. Ova studija je uključila 13 grupa po 6 životinja (mužjaci, tjelesne mase 150-200 g) . Pentadekapeptid BPC 157 je davan svakodnevno, u vodi za piće ili per os ad libitum u dvjema dozama (10 μg/kg ili 10 ng/kg), u kotretmanu od 1. do 29. dana ili pak u posttretmanu, u kojem se za početak tretmana se čekao 15. dan i uspostava plućne hipertenzije, te su ga životinje primale od 15. dana do 29. dana. U kontrolnim skupinama dokazana je izrazita hipertrofija desne klijetke, a histološki je bilo vidljivo masivno zadebljanje medije prekapilarnih arterija, uz kliničko pogoršanje zbog plućne hipertenzije tijekom 4. tjedna nakon injekcije MCT te smrtnošću među pokusnim životinjama uslijed desnostranog srčanog zatajivanja. U kotretmanu pentadekapeptid BPC 157 je u svim modalitetima primjene učinkovito spriječio nastanak plućne hipertenzije sa svim manifestacijama, dok je u posttretmanu pentadekapeptid BPC 157 isto tako u svim modalitetima primjene uspješno sanirao već razvijenu PAH. Zaključujemo da pentadekapeptid BPC 157 uspješno prevenira i terapira MCT induciranu plućnu hipertenziju i plućno srce u štakora.
