Abstract

Background. Spinal cord injury is devastating lesion with life time disability. Previously pentadecapeptide BPC 157 exhibited a healing effect on brain traumas. Aim. To verify influence of pentadecapeptide BPC 157 on rat spinal cord injury. Material and methods. Deeply anaesthetized Wistar Albino male rats were subjected to surgery with injury: after laminectomy L2-L3 60 second compression with neurosurgical piston (60-66 g) on exposed dural sac. One single medication (IP) (saline 5ml/kg; pentadekapeptide BPC 157 200ug/kg; 2 ug/kg) was 10 min post-injury and sacrifice was at 7, 15, 30, 90, 180 and 360 days. Spinal cord injury was followed with tail motor function, histology of spinal cord and tail and electromyography. Results. BPC 157 rats have significantly higher motor score and in all followed time and less spasticity from 4 weeks till the end. From 2 weeks BPC 157 rats have less grey matter edema and less cyst and axonal necrosis in white matter of spinal cord. All treatment groups show increased number of small axons starting from 2 weeks witch is unexpected finding. Conclusion. Given only once immediately after spinal cord injury, pentadecapeptide BPC 157 show strong neuroprotective effect. Further studies with markedly postponed administration of BPC 157 are required.

Abstract in Croatian

Uvod. Ozljeda kralježnične moždine je teška tjelena ozljeda s trajnim invaliditetom. Dosadašnja istraživanja potvrdila su pozitivan učinak pentadekapeptida BPC 157 na traumatsku ozljedu mozga. Cilj. Utvrđivanje učinka pentadekapeptida BPC 157 na neurološko oštećenje kod ozljede kralježnične moždine. Materijali i metode. U duboko anesteziranih muških Wistar Albino štakora nakon laminektomije L2-L3 tehnikom kompresije neurovaskularnom klipsom 60-66 g trajanja 60 s učinjena je ozljeda sakrokokcigealne kralježnične moždine. BPC 157 je primjenjen samo jednom - 10 minuta nakon ozljede sakrokokcigealne KM, u dozi 2 μg/kg ili 200 μg/kg, a NaCl 5 ml/kg. Životinje su žrtvovane 7, 15, 30, 90, 180 i 360 dana nakon ozljede. Praćena je motorika i spasticitet repa, histopatologija moždine i repa i elektromiografija repa. Rezultati. BPC 157 je pokazao bolji i brži neurološki oporavak motorne funkcije repa i manji spasticitet repa. Histopatologijom moždine nalazi se manje edema u sivoj tvari moždine, te manje cističnih promijena i manje gubitaka aksona u bijeloj tvari moždine od 15. dana do kraja praćenja. Histopatologijom repa potvrđen je povećan broj malih aksona u svim skupinama, što je neočekivani rezultat. Zaključak. BPC 157 primjenjen u perakutnoj fazi sekundarne ozljede kralježnične moždine pokazuje značajan neuroprotektivni učinak. Planiran je nastavak istraživanja s primjenom BPC 157 u kasnijoj fazi sekundarne ozljede.
