Abstract

Cutaneous squamous cell carcinoma (cSCC) is the second most common skin cancer. HER pathways play a key role in cancer biology, their dysregulation leads to growth and spread of tumour cells. Main hypothesis of the study was: higher expression of transmembrane receptors of tyrosine kynase (EGFR, HER2 and HER3), proliferation marker Ki67 and protein p53 is expected in cSCC compared to normal skin, and in multiple tumours. Primary objective of our investigation was to compare expression of EGFR, HER2 and HER3, proliferation marker Ki67 and protein p53 in cSCC and normal skin. Investigated group included 101 patient with 134 cSCC of different patohistology stage according to Broders classification. Control group included 134 samples of normal skin of the same patients analyzed in investigated group. Results of our investigation showed significantly higher expression of EGFR receptors, Ki67 and protein p53 in tumour compared to normal skin, as well as in multiple compared to solitar tumours. The above mentioned results led to conclusions that EGFR, Ki67 and p53 have a key role in cSCC development and that they are related to malignant phenotype. Results of this study could lead to EGFR inhibitors implementation in treatment of patients with aggressive cSCC.

Abstract in Croatian

Planocelularni karcinom kože (cSCC) drugi je po učestalosti maligni tumor epitela kože i sluznica. HER signalni putevi igraju važnu ulogu u biologiji nastanka raka, njihova deregulacija dovodi do rasta i rasapa tumorskih stanica. Osnovna hipoteza rada bila je: očekuje se veća izraženost transmembranskih receptora tirozin kinaze (EGFR, HER2 i HER3), biljega Ki67 i proteina p53 u cSCC u odnosu na zdravu kožu, te u osoba s multiplim tumorima. Primarni cilj našeg rada bio je usporediti izraženost EGFR, HER2 i HER3, biljega Ki67 i proteina p53 u cSCC u odnosu na zdravu kožu. Ispitivana skupina je obuhvatila 101 bolesnika kod kojih je utvrđeno 134 planocelularnih karcinoma kože, različitog patohistološkog stupnja određenog prema Brodersovoj klasifikaciji. Kontrolnu skupinu činila su 134 uzorka zdrave peritumorske kože istih bolesnika, obrađenih u ispitivanoj skupini. Rezultati našeg istraživanja pokazali su statistički značajno veću izraženost receptora EGFR, biljega proliferacije Ki67 i proteina p53 u tumorskoj u odnosu na zdravu kožu, kao i u multiplim tumorima u odnosu na solitarne. Možemo zaključiti da EGFR, Ki67 i p53 imaju važnu ulogu u nastanku cSCC, te su povezani s malignijim fenotipom. Rezultati ovog istraživanja mogli bi imati kliničke implikacije u smislu praktične primjene inhibitora EGFR u bolesnika s agresivnim oblicima cSCC.
