Utjecaj interakcija gena i okolišnih čimbenika na pojavnost metaboličkoga sindroma.

Abstract
The role of the gene environment interactions in the pathogenesis of metabolic syndrome (MetS) has not been fully clarified yet. PPARγ regulates the expression of key genes involved in adipogenesis, lipid and glucose metabolism, inflammation and immunity. LPL, IL6, ACE and AT1R belong to different PPARγ target genes. The aim of the study was to estimate the influence of interactions between PPARγ gene variants, its target genes and diet on MetS and its traits. The study included 527 participants (263 with MetSy and 264 controls). Genotyping of PPARγ Pro12Ala, LPL PvuII (-/+), IL6 -174G>C, ACE I/D and AT1R 1166A>C was performed using polymerase chain reaction based methods.
In the group of participants with LPL PvuII (-/-), those with PPARγ Pro12Ala or Ala12Ala genotype had lower odds for MetSy. PPARγ Ala12 variant was found protective in patients with IL6 -174GG genotype, while in the case of IL6 -174C allele carriers, for PPARγ Ala12 carriers larger odds for high glucose levels compared to Pro12 variant were observed. Interactions of PPARγ and ACE were significant for BMI. In the group with ACE DD genotype, those with PPARγ Pro12Ala or Ala12Ala genotype had greater odds for obesity.
Mediterranean or mixed diet was more prevalent in the group of 1166AC or CC genotype carriers.

U patogenezi metaboličkoga sindroma (MetS) nije još sasvim razjašnjena uloga interakcija gena i okoliša. PPARγ regulira ekspresiju ključnih gena u adipogenezi, metabolizmu masti i šećera, upali i imunosti. Među različite ciljne gene PPARγ pripadaju i LPL, IL6, ACE i AT1R. Cilj ove studije bio je ispitati interakcije između varijanti PPARγ i njegovih ciljnih gena, te prehrane i njihovu povezanost s MetS i pojedinim njegovim sastavnicama.
Istraživanje je provedeno na 527 ispitanika (263 s MetS i 264 kontrole). Genotipizacija PPARγ Pro12Ala, LPL PvuII (-/+), IL6 -174G>C, ACE I/D i AT1R 1166A>C provedena je metodama temeljenim na lančanoj reakciji polimerazom.
U skupini s genotipom LPL PvuII (-/-), nositelji genotipa PPARγ Pro12Ala ili Ala12Ala su imali manji rizik za razvoj MetS u odnosu na ispitanike s genotipom Pro12Pro. Varijanta gena PPARγ Ala12 ima zaštitnu ulogu u ispitanika s genotipom IL6 -174GG, dok je u nositelja alela IL6 -174C i alela PPARγ Ala12 zabilježen veći rizik razvoja povišenih koncentracija glukoze na tašte u usporedbi s nositeljima varijante PPARγ Pro12. Interakcija između PPARγ i ACE je bila značajna za vrijednosti BMI. U skupini ispitanika s genotipom ACE DD, nositelji genotipa PPARγ Pro12Ala ili Ala12Ala imali su veći rizik za razvoj debljine. Mediteranska ili mješovita prehrana bila je učestalija kod nositelja genotipa AT1R 1166AC ili CC u odnosu na genotip AT1R 1166AA.
